Ipamorelin is a small synthetic peptide that prompts the pituitary gland to release a pulse of growth hormone. It is popular with beginners because, unlike older peptides in its class, it barely affects cortisol, prolactin or appetite.
- It mimics ghrelin, the “hunger hormone”, at the growth hormone receptor.
- Effects are gradual: sleep, recovery and body composition over months.
- It is a research compound in Canada, not an approved medicine.
Ask in any Canadian gym about peptides and ipamorelin usually comes up first. Its reputation as the “gentle” growth hormone peptide is mostly deserved, but the marketing around it often promises more than the research shows. This guide covers what ipamorelin does, what it does not do, and the practical points to understand before handling it.
What ipamorelin is
Ipamorelin is a pentapeptide, a chain of five amino acids, developed by Novo Nordisk in the 1990s. It belongs to a group called growth hormone secretagogues (GHS): compounds that make your own pituitary release more growth hormone (GH) rather than supplying GH directly. It acts on the GHS receptor, the same receptor that ghrelin uses.
Once GH is released, the liver responds by making more IGF-1, which carries out many of GH’s effects on muscle, bone, connective tissue and fat metabolism.
Why it is called “gentle”
Earlier secretagogues such as GHRP-6 and GHRP-2 also raise GH, but they tend to raise cortisol and prolactin too, and GHRP-6 in particular causes strong hunger. In the original animal research, ipamorelin raised GH with little or no change in cortisol or ACTH, even at high doses. That selectivity is the main reason it is chosen over its relatives.
| Peptide | GH release | Hunger | Cortisol and prolactin |
|---|---|---|---|
| Ipamorelin | Moderate | Mild | Little change |
| GHRP-2 | Strong | Moderate | Can rise |
| GHRP-6 | Strong | Strong | Can rise |
| MK-677 (oral) | Moderate, all day | Strong | Small rise |
What the research shows
Human data on ipamorelin is limited. It reached clinical trials as an intravenous treatment to speed bowel recovery after abdominal surgery, where it was well tolerated but did not beat placebo clearly enough to be approved. There are no long-term trials in healthy people looking at muscle, fat loss or ageing.
What we can say from growth hormone research in general: raising GH in adults tends to improve sleep depth, skin and connective tissue, and lean-to-fat ratio slowly over months. In healthy young people the effect on muscle size and strength is small. Expect ipamorelin to be subtler than injected growth hormone, because a secretagogue only amplifies your natural pulses.
Ipamorelin with CJC-1295
Ipamorelin is often paired with a growth hormone-releasing hormone (GHRH) analogue such as CJC-1295 or sermorelin. The two work on different receptors: GHRH tells the pituitary to release GH, and the ghrelin mimic amplifies that signal and reduces the brake (somatostatin). Together they produce a larger pulse than either alone. Our guide to CJC-1295 and ipamorelin explains the combination in more detail.
Side effects
- Redness or itching at the injection site
- A warm flush or light-headedness shortly after injecting
- Mild water retention and, with ongoing use, tingling or numbness in the hands
- Headache in some users
- Higher fasting blood sugar over time, as with anything that raises GH
Because GH and IGF-1 promote cell growth and raise blood sugar, ipamorelin is not a good idea for anyone with active cancer, a history of cancer, diabetes or diabetic eye disease. Checking IGF-1 and fasting glucose or HbA1c at LifeLabs or Dynacare before and during use shows whether it is doing anything and whether blood sugar is drifting. Our bloodwork guide explains how to order tests in Canada.
Timing and food
GH release is blunted by high blood sugar and insulin. That is why secretagogues are usually discussed alongside an empty stomach: before bed at least two hours after eating, or first thing in the morning before breakfast. Your largest natural GH pulse comes in the first hours of deep sleep, which is why bedtime is the most common choice.
Handling and storage
Ipamorelin is supplied as freeze-dried powder. It is mixed with bacteriostatic water and given under the skin with an insulin syringe. Our step-by-step guide to reconstituting peptides covers sterile technique and the concentration maths, and the Vita Ipamorelin 5 mg page includes a measuring calculator and an injection-site guide.
- Powder: cool, dry and dark; the fridge is best for longer storage.
- Mixed: always refrigerate at 2–8 °C and use within about four weeks.
- Never freeze a mixed vial or leave it in a hot car in summer.
Legal status in Canada
Ipamorelin is not a controlled substance under the Controlled Drugs and Substances Act, but it is not approved by Health Canada for human use, and it is sold for research. It is on the World Anti-Doping Agency prohibited list, so tested athletes in Canadian sport (including university athletes under the CCES) must not use it.
Frequently asked questions
Is ipamorelin the same as growth hormone?
No. It makes your pituitary release more of your own growth hormone in pulses. Injected growth hormone (somatropin) supplies the hormone directly and has much larger effects.
How long does ipamorelin take to work?
Sleep changes may be noticed within weeks. Changes in body composition, skin and recovery, if any, take months.
Does ipamorelin make you hungry?
Much less than GHRP-6 or MK-677. Some people notice mild hunger after injecting, but most do not.
